Abstract

Penelitian dilakukan dengan tujuan: (1)  mengaklimasi beberapa jenis ikan hasil tangkapan dari Sungai Serayu Banyumas pada kolam percobaan (ex situ); (2) mengetahui profil hormonal estrogen (17ß-estradiol), progesteron, testoteron, FSH ikan jantan dan betina selama satu siklus reproduksi; (3) mengetahui gambaran histologis perkembangan testis dan ovarium ikan tersebut selama satu iklus reproduksi; (4) mengetahui beberapa aspek fisiologis dan ekologis (karakteristik substrat habitat baru, kebiasaan pakan); (5) natural spawning (pemijahan alami di kolam budidaya) dibantu dengan metode induksi gonadotrophin analogue. Preparasi sediaan histiologis dengan metode paraffin, pewarnaan Haematoxylin Eosin; analisis hormon dengan metode ELISA menggunakan mesin Microplate Reader-LB-6200 Labotron. Uji kesukaan pakan dari media/substrat pemeliharaan). Hasil yang diperoleh: Ikan Brek dan Lukas sudah berhasil adaptif (data indeks kesukaan pakan)  masing-masing 56,893%  dan 62,854%. profil hormon sampai akhir periode predomestikasi 8 bulan, berurutan dari Brek dan Lukas : (17ß-estradiol = 1058,06–854,85 pg./ml; 395,18–854,85pg/ml); progesteron = 0,29–0,72 ng/ml; 0,20-0,52 ng/ml); (FSH = 10,87-15,68 mlU/ml; 8,26-13,74 mlU/ml); (testosteron = 4,18-9,92 ng/ml; 3,98-9,34 ng/ml). ada 5 (lima) tahapan perkembangan oogenesis Brek dan Lukas sama yakni chromatin nucleolar stage (cns); perinucleolar stage (ps); cortical alveolar stage (cas); vitellogenic stage (vs) dan mature/ripe stage (ms). Tahapan spermatogenesis Brek dan Lukas sama yakni tahap spermatogonium; spermatosit primer; spermatosit sekunder; spermatid, spermatozoa. Pada predomestikasi 4 bulan hanya induk Lukas yang mampu memijah; dan pada akhir periode predomestikasi 8 bulan, Brek dan Lukas berhasil memijah. Semua induk stok yang masih berada di kolam akan terus didomestikasi untuk menjadi stok induk super suatu saat. Larva pasca penetasan tetap dipelihara untuk penelitian lebih lanjut.
 
This research conducted with the aim of: (1) the acclimation of several fish species from Serayu river Banyumas in the experiments pond (ex situ); (2) knowing the estrogen hormonal profile (17ß-estradiol), progesteron, testosteron, FSH, male and female fish during pre-domestication period/a reproductive cycle; (3) knowing histological picture of the testis and ovary development of these fish during one reproductive cycle; (4) identified several ecological aspects (substrate characteristics of new habitats, feeding habits); (5) natural spawning (naturally spawning in the cultivation pond) assisted with analogue GnRH induction methods. Preparation of histologists supply with paraffin method, Haematoxylin Eosin staining; hormon analysis by ELISA method using a machine-Microplate Reader LB-6200 Labotron. Test of the media's/substrate maintenance favorite food (index of electivity, and index of preponderance. The result: Brek and Lukas had managed to adaptive (feed preference index / index of preponderance 56.893and 62.854, respectively). Hormone profiles until the end of 8 months pre-domestication period, from Brek and Lukas, respectively: (17ß-estradiol = 1058,06-854,85 pg/ml; 395,18-854,85pg/ml); progesteron = 0,29-0,72 ng/ml; 0,20-0,52 ng/ml); (FSH = 10,87-15,68 mlU/ml; 8,26-13,74 mlU / ml); (testosteron = 4,18-9,92 ng/ml; 3,98-9,34 ng/ml). There are 5 (five) stages of oogenesis development Brek and, Lukas as the chromatin nucleolar stage (cns); perinucleolar stage (ps); cortical alveolar stage (cas); vitellogenic stage (v) and mature / Ripe stage (ms). Spermatogenesis stages of Brek, and Lukas is same that are spermatogonia stage; primary spermatocytes; secondary spermatocytes; spermatids, spermatozoa. At 4 months pre-domestication only Lukas parents  that is capable to spawn; and at the end of the 8 months pre-domestication period, Brek and Lukas managed to spawn. All of the parents stock which is still in the pond will continue to be domesticated to make super parents stock someday. Post-hatching larvas remain preserved for further research
